What is Elisa washer?

Elisa washer is specially designed for cleaning enzyme standard plates and is usually used in conjunction with an enzyme standardizer. It is mainly used to clean some residues after the enzyme plate assay, thus reducing the errors caused by residues in the subsequent assay process. Elisa plate washer has been widely used in hospitals, blood stations, health prevention stations, reagent factories, and research laboratories for enzyme plate cleaning.

Automatic Elisa plate washer is an essential instrument in multi-phase ELISA experiments. It is mainly used to wash some residual materials after enzyme plate assay, and wash to remove unbound antigens and impurities, after washing, only specific antibodies are left on the solid phase carrier. The impurities in other immunoglobulins and serum are washed away during the washing process because they cannot bind to the solid-phase antigen. This reduces the error caused by residues in the subsequent assay process, and also automatically flushes the pipeline to prevent blockage caused by crystallization of the wash solution during reuse.

Elisa washer is mainly used in the process of ELISA experiments. Its function is to wash the unbound antigen or antibody and impurities so that only the antigen-antibody complex formed by specific binding is retained on the solid phase carrier. Therefore, Elisa plate washers are usually used in conjunction with ELISA, and this washing step can affect the accuracy of subsequent assay results.

a. Although the Elisa washer is usually more robust and durable, there are certain requirements for the working environment: the plate washer needs to be placed on a clean, horizontal work surface with a large enough operating space, the operating voltage needs to be kept stable during operation, the ambient temperature should be between 15°C and 40°C, the ambient humidity between 15% and 85%, and the air in the operating environment should be kept clean to avoid water vapor, smoke, and dust, which are basically as the general instrument. These requirements are basic to the general working environment of the instrument.

b. Before using the Elisa plate washer, you need to check whether the waste liquid bottle is empty, whether there is enough wash liquid in the bottle, whether the connection of each part is tight, and whether the internal and external hose liquid circuit should be kept free of knots, extrusion, and leakage, and should not be in contact with sharp objects to prevent breakage. If any liquid circuit needs to be replaced, the interface should be carefully screwed down and replaced with a new liquid circuit.

c. In the process of use, the waste liquid should be dumped in time (when the waste liquid reaches 2/3 of the bottle height, you can dump the waste liquid), so that the failure of the Elisa washer caused by the backflow of the waste liquid due to the overfilling of the bottle. Put the bottle back to its original place after the waste solution is poured out, and make sure the seal between the bottle and the cap. Note that the waste liquid should be handled as biohazardous material.

d. At the end of the work of Elisa microplate washer every day, clean the inside and outside of it, and wipe the outer wall of the plate washer and the load table with a wet rag to ensure its cleanliness. Clean all the pipes with distilled water to prevent the crystals of the washing solution from blocking the pipes. Turn off the power and unplug it in time after use.

e. If the residue is too much, it may penetrate the rumen, which will cause damage to the appearance or performance of the instrument, so the bench and the exposed parts of the instrument need to be cleaned. Note that organic solvents, grease, or corrosive liquids should not be used to clean the instrument.

f. A particularly important point is the cleaning and maintenance of the wash head. Wash head is an important part of the Elisa washing machine, which directly affects the main performance of the washer, so the maintenance of the wash head is the focus of the entire instrument maintenance. The accumulation of suspended particles or crystals in the washing solution for a long time will block the fine pipes in the washing head, so please use a fresh washing solution and check frequently for poor flow or blockage of the liquid.

g. When using the Elisa microplate washer frequently, please start the flushing program frequently and clean the pipeline to ensure the pipeline is clear. When the wash head is found to have poor liquid flow or blocked pipes, please turn off the machine and remove the wash head to unclog the corresponding suction/injection needles (coarse needles are used to unclog the suction tube and fine needles are used to unclog the injection tube), after unclogging, put the wash head back into the instrument and turn on the machine to execute the rinsing program several times until normal.

Common faults and troubleshooting of Elisa washer

A. Elisa washing machine cannot wash the plate during the pressure alarm

Elisa washer is composed of two parts of the head and body for plugging, due to the vibration of the machine, will cause the head and the body connection of the electrical socket, the pipe interface to poor contact, the air pump not to work, and generate pressure alarms.

The treatment method is to adjust the position of the head and body, if necessary, the head and body are completely separated and re-plugged to ensure good contact at the connection, and the air pump can work normally.

If there is still a pressure alarm after the air pump works, check whether there is air leakage at the pipeline connection of the machine, especially pay attention to the pipeline alignment on the cap of the liquid wash after replacing the liquid wash, and then tighten the cap. After eliminating the air leakage phenomenon, the machine can work normally.

B. Incomplete cleaning, leaving a residual liquid

This phenomenon is caused by impurities blocked within the cleaning head. You can use the machine's special filament through the needle, and the cleaning head one by one to remove impurities so that it can be smooth.

C. In and out of the water control failure, cleaning head out of water non-stop

The water in and out of the cleaning head is controlled by the solenoid valve to squeeze or relax the water hose. Due to the long time squeezing, the hose will be aging and elasticity will be reduced, so that when the hose is clamped, the washing liquid will still pass through and the water will not stop coming out of the cleaning head. At this time, you can replace the new hose. If there is no new hose, the hose can be pulled forward or backward by hand to change the contact position of the solenoid valve and hose, so that it can normally control the water in and out.

D. The lifting action of the cleaning head is not flexible

The lifting and lowering of the washboard head are controlled by the lifting and lowering of the control lever on the lower left side of the washboard. The gap between the control rod and the sleeve is very small, due to the entry of leakage or impurities, the control rod and the sleeve will produce adhesion, resulting in inflexible movement. Remove the impurities between the rod and the sleeve, move the control rod up and down several times by hand, and then drop a little lubricant to make the control rod move freely, so the washboard head can be lifted normally.

E. Inaccurate washboard position

The change of the wash plate position is controlled by the backward and forward parallel movement of the specimen dragging plate on the machine plate. If the dragging plate is not flexible and not accurately in place, the cleaning head cannot be accurately inserted into the specimen pool, making the wash liquid outflow and unable to wash the plate normally. On the reverse side of the dragging board inlaid with two small circular magnets, due to long-term erosion of the liquid, the magnets will become rough and uneven due to corrosion, so that the movement of the dragging board is blocked, and cannot be accurately in place. The solution is to remove the dragging board, use sandpaper to smooth out the rough magnets, and use a soft cloth to clean the reverse side of the dragging board and the body where the dragging board is placed so that the two planes in contact with the machine are smooth and flat, and the dragging board moves freely and can be put in place accurately.
